Novel reproducible genetic associations, disease overlaps, clinical studies, actively protective biology, and repurposing opportunities 

PrecisionLife presented its ME/CFS and long COVID research progress at the 2026 International ME Conference highlighting major advances in the biological understanding of ME/CFS and long COVID, including novel reproducible genetic associations, shared disease mechanisms, patient stratification approaches, genetic testing, and precision drug repurposing opportunities. 

Using the PrecisionLife combinatorial analytics platform, researchers analyzed large-scale genomic datasets from UK Biobank, DecodeME, Sano GOLD, and the US All of Us Research Program to identify complex combinations of genetic variants associated with ME/CFS and long COVID. These analyses uncovered thousands of disease-associated genetic signatures and demonstrated significant biological overlap between the two conditions.

Key findings:

  • Identification of 7,555 SNPs linked to ME/CFS across DecodeME and UK Biobank cohorts, mapped to 259 candidate core genes and 2,052 peripheral genes.

  • Discovery that 76 genes associated with long COVID were also significantly associated with ME/CFS, supporting substantial shared biology between the conditions.

  • Independent validation of long COVID disease signatures across UK and US cohorts, with up to 94% gene reproducibility observed in the All of Us population.

  • Identification of enriched mechanisms linked to neurological dysregulation, inflammation, cellular stress responses, calcium signaling, and neurotransmission.

  • Development of novel non-invasive genetic tests designed to help patients better understand the biological mechanisms linked to their symptoms using saliva or cheek swab samples.

  • Discovery of multiple drug repurposing opportunities, including genetically supported therapeutic targets and biomarkers that could help identify patients more likely to benefit from GLP-1 receptor agonists or other therapies.

  • New research into actively protective biology” identifying potential resilience mechanisms that may help protect some individuals from developing ME/CFS despite elevated disease risk.

Precision medicine for complex chronic diseases  

The findings reinforce growing evidence that ME/CFS and long COVID are highly polygenic, multisystem diseases involving multiple overlapping biological mechanisms. PrecisionLife’s mechanistic patient stratification approach aims to support the development of more targeted diagnostics, prognostic tools, clinical trial designs, and precision therapies for patients with complex chronic diseases.

The poster also highlights ongoing work with Ovation to analyze genomic and longitudinal clinical data from 25,000 GLP-1 treated patients, with the goal of identifying biomarkers that predict treatment response and side effect risk.
